SmartRNAs: engineered guide RNAs; inspired by the natural role of microRNAs, which evolved to regulate entire gene networks rather than single genes. • Building on recent advances in microRNA biology, including insights into RISC structure and kinetic binding dynamics, we developed a second-generation algorithm that designs smartRNAs capable of engaging multiple intended and optional targets, while actively excluding predefined off-targets. • SmartRNAs are designed for efficacy, specificity, and deliverability, providing a scalable and programmable platform for reprogramming cellular behavior at the systems level.
